Overview
The 4-alkoxy-substituted benzyl moiety (Wang) linker is commonly used for Fmoc chemistry. This linker is stable to basic conditions, but cleaved smoothly under acidic conditions with ≥ 90% trifluoroacetic acid (TFA) to furnish a peptide acid. Coupling of the first amino acid to the linker is typically done with 4-(Dimethylamino)pyridine (Dmap) as a catalyst. This step is subject to unwanted side reactions and can be problematic. For convenience, Wang resins are typically supplied pre-loaded with the first amino acid.
