Overview
The 4-alkoxy-substituted benzyl moiety (Wang) linker is commonly used for Fmoc chemistry. This linker is stable to basic conditions, but cleaved smoothly under acidic conditions with ≥ 90% trifluoroacetic acid (TFA) to furnish a peptide acid. Coupling of the first amino acid to the linker is typically done with 4-(Dimethylamino)pyridine (Dmap) as a catalyst. This step is subject to unwanted side reactions and can be problematic. For convenience, Wang resins are typically supplied pre-loaded with the first amino acid.

Modern SPPS is most effectively utilized with instrumentation to automate and accelerate the reaction process. The resin can be loaded into the instrument along with a bank of amino acids in solution and reactions can run autonomously. Microwave heating can be used to dramatically increase the rate and purity of the reaction.
