Overview
PEG-PS resin cores are ideal for longer and more challenging peptides. The PEG-PS technology incorporates the benefits of PEG into a PS core to provide a balance between swelling and polarity. In addition, these hybrid resins maintain the mechanical integrity of polystyrene with the reduced aggregation effects of PEG. ProTide® PEG-PS resins are optimized to create an environment perfect for synthesizing long or difficult peptides.
Linker selection is an additionally important factor for successful synthesis. Serving as the connection point for the first amino acid in the sequence, linkers typically furnish C-terminal acids or amides upon cleavage. Commonly used C-terminal acid linkers include the robust Wang linker and the hyper acid-sensitive chlorotrityl-type linker, while the Rink Amide linker is a commonly employed C-terminal amide linker. For C-terminal peptide acids, the first amino acid coupling can be difficult and prone to epimerization, so resins are often conveniently preloaded with the first amino acid.
